The aim of this work was to investigate the effect of anaesthesia with AIA (350 mg/kg) or Imidazole (400 mg/kg) received a single dose of Enflurane or Isoflurane (1 mL/kg). In AIA treated mice, total CYP levels were strikingly diminished when the xenobiotic was administered either alone or plus the anaesthetics. In Imidazole treated mice, total CYP levels were first induced but they returned to control values by effect of the anaesthesia. CYP2E1 activity was strikingly increased in mice receiving the anaesthetics being higher in the group treated with Enflurane and Imidazole: this fact would indicate an increase in Enflurane metabolism. In AIA treated animals, anaesthesia produced no additional effect. Although, in those animals receiving Imidazole, Phase II enzymes activities were less reduced by the action of Isoflurane. A diminishment of Tryptophan pyrrolase activity was observed when the anaesthetics were administered to the Imidazole group, indicating a reduction in the heme regulatory pool. In conclusion, the effects of Enflurane and Isoflurane on drug metabolizing system were dependent on the CYP status and the anaesthetic assayed in spite of the fact that both compounds have a similar chemical structure.
