Milk fat composition is one of the factors influencing the nutritional quality of dairy products. We studied the A293V polymorphisms in the Stearoyl-Coenzyme A Desaturase locus (SCD) and the K232A polymorphisms in the Acyl CoA: Diacylglycerol Acyltransferase 1 locus (DGAT1). Both enzymes have an important role in lipid metabolism. The aims of the present study was to determine gene frequencies for both loci in the breed Siboney de Cuba (N=165). The methodology for both loci was amplification created restriction sites (ACRS). We observed that the SCDA allele and the DGAT1A allele were the most frequent in the studied population. Three genotypes were identified at each locus, with the following frequencies: 0.821, 0.172 and 0.007 for genotypes AA, AS and SS, respectively, at the SCD locus and 0.686, 0.212 and 0.102 for genotypes AA, AK and KK, respectively, at the DGAT1 locus. The SCD locus was in Hardy-Weinberg equilibrium, whereas a deviation from the Hardy-Weinberg equilibrium (p <0,001) was found at the DGAT1 locus.
